Net peptide content
The fraction of a lyophilised powder's mass that is actually peptide, after subtracting residual water and counterion — a different measurement from chromatographic purity, and reported far less often.
Lyophilised peptide is never pure peptide by weight. The powder in a vial also contains residual water and the counterion carried over from purification — commonly trifluoroacetate or acetate — which together can account for a noticeable fraction of the mass. Net peptide content is the measurement that states what fraction of the powder is peptide, typically determined by amino-acid analysis or nitrogen determination.
It is not the same number as HPLC purity, and the two are routinely conflated. Chromatographic purity compares the target peak with other detected peaks and can read 99% while the powder is only 80–85% peptide by mass. Purity describes the quality of the peptide fraction; net content describes how large that fraction is. A vial's label mass is the gross powder weight unless the certificate states otherwise.
The distinction matters wherever mass enters a calculation, because a concentration computed from gross powder weight overstates the peptide actually present. Certificates that report net content alongside purity resolve the ambiguity; most report purity alone, which is why the distinction belongs in any careful reading of a COA.
