The composition draws on several distinct literatures. Arginine is the direct substrate for nitric oxide synthase, while citrulline is described in clinical pharmacology reviews as a more efficient route to raising plasma arginine because it bypasses hepatic first-pass metabolism and intestinal arginase; ornithine sits in the same urea-cycle loop that regenerates arginine from citrulline. Taurine is one of the most abundant free amino acids in skeletal muscle and heart, where the experimental literature associates it with membrane stabilisation, osmoregulation, and modulation of vascular tone. L-carnitine is the obligatory carrier shuttling long-chain fatty acids across the inner mitochondrial membrane for beta-oxidation.
N-acetylcysteine functions in this context as a cysteine donor for glutathione synthesis, and controlled exercise studies have used it to probe the role of redox status in skeletal-muscle fatigue. Glutamine, lysine, and proline are studied primarily as substrates in protein and collagen synthesis rather than as signalling molecules.
